| I. Background | |

CLEC5A（C-type lectin domain family 5 member A）是位于人类 12 号染色体（12q24.31） 的Ⅱ型跨膜糖蛋白基因，属 C 型凝集素受体家族，广泛表达于髓系细胞（巨噬细胞、中性 粒细胞）、内皮细胞及肿瘤细胞。作为模式识别受体，其与 TLR2 协同激活先天免疫，促 进中性粒细胞胞外陷阱（NET）形成以清除细菌（如李斯特菌）和病毒（如登革热、 H5N1），但过度激活会引发细胞因子风暴，导致脓毒症、登革热休克综合征等致命性炎症 反应，并与红斑狼疮、炎症性肠病过度炎症损伤相关。在肿瘤中，CLEC5A 功能双面：在 白血病、胃癌、胶质瘤中顺利获得激活 AKT/mTOR 通路促进增殖、抑制凋亡，驱动肿瘤进展； 在骨肉瘤中则抑制钙调磷酸酶/NFATc1 通路减少转移，呈现抑癌特性。 |
